He developed an interest in politics when he was pursuing his graduation in 1984.

Before entering politics, he started his career as a journalist in New Delhi in 1986. Two years later, he joined the Jan Satta (Sister publication of The Indian Express) and moved to Mumbai.

Nirupam resigned from the Shiv Sena in March 2005; after a long rivalry with the BJP leader and the former Telecommunications Minister Pramod Mahajan, whom he had accused of corruption.

He joined the Indian National Congress (INC) in April 2005 and was soon appointed as the General Secretary of the Maharashtra Pradesh Congress Committee (MPCC) by Sonia Gandhi.

He was elected to the 15th Lok Sabha in 2009 from the North Mumbai constituency but lost the seat to Gopal Shetty in 2014.

He has been organizing International Wrestling Tournaments every year for the past 10 years with the top-ranked wrestlers.

In October 2016, Sanjay had questioned the government and asked for proof for the surgical strikes, for which he was criticized by all including his own party. Sanjay even got a lot of death threats. His wife, Geeta, wrote an open letter to Prime Minister Narendra Modi asking for protection for her family.

In September 2018, government schools were showing a documentary about Prime Minister Narendra Modi on which Sanjay remarked that schools should not be showing a documentary about an illiterate person such as Narendra Modi. In 2019, members of the Mumbai Congress had criticized his leadership as the President of the Mumbai Congress. They had a meeting with Mallikarjun Kharge (Maharashtra Congress Committee in-charge) and demanded Nirupam’s replacement. On 25 March 2019, he was replaced as the President of the Mumbai Congress, by Milind Deora. Sanjay was named the Congress party’s candidate of the North West Mumbai Constituency for the 2019 Lok Sabha Elections.
