- Sherfane Rutherford started playing cricket at the young age of six for the village team of Enmore.
- He made his first-class debut on 15 April 2017 for Guyana against the Windward Islands at Providence, Rhode Island, New England in the 2016–17 Regional Four-Day Competition.
- In June 2018, he got selected to play for West Indies B in the Global T20 Canada tournament. He was the highest runscorer for West Indies B and scored 230 runs in eight matches.
- In the 2018 T10 Cricket League, he scored 46 runs off 21 balls for Bengal Tigers.
- He scored 2 runs off 6 deliveries in his first T20I match for West Indies against Bangladesh on 22 December 2018.
- In December 2018, Delhi Capitals acquired him for Rs 2 crore in the 2019 Indian Premier League (IPL) auction.
- In 2019, the IPL franchise Mumbai Indians (MI) traded Mayank Markande to Delhi Capitals (DC) for Rutherford.
- In a 2020 Pakistan Super League (PSL) match for Karachi Kings, he was spotted wearing gloves of the IPL franchise Mumbai Indians (MI) after which many fans criticised him for not wearing Kings’ gloves.
